I totally agree with you on using keywords in your URL but the only problem is that this strategy cannot be used on existing domains.
I think one should consider SEO planning when one plans to design a website.

Be consistent in linking behavior. If you link to homepage.com, always link to homepage.com, and not to homepage.com, homepage.com/index.php and homepage.com/index.php&id=123.

Yes, it does affects one ranking for a keyword because SE will first of all recognize that URL for having its keyword on its name.
So it is good to have a keyword optimized domain name.

Nice question.... URL is the most important SEO on page element. All search engines are looking first URL and after this title, meta data and content. So the most important keyword must take place in your URL.
